Set of reagents for species identification of sturgeon fish.
The kit is designed for molecular genetic analysis of sturgeon fish (Acipenseridae) for the purpose of species identification and identification of individuals of hybrid origin.
The COrDIS SturGene ID kit is a test system, consisting of two PCR modules, based on
– multiplex PCR amplification of nuclear DNA using fluorescently labeled primers, specific to five highly polymorphic microsatellite loci: Afug41, Afug51, An20, AoxD161, AoxD165.
– PCR amplification using species-specific primers for the control region of mitochondrial DNA.
Analysis of five microsatellite loci allows us to establish the species, and also identify an individual of hybrid origin. In the latter case, mtDNA analysis makes it possible to determine the species identity of the hybrid along the maternal line, and the origin of the hybrid on the paternal line is determined by the presence of alleles in the hybrid, not characteristic of the maternal species.
The kit was developed based on the method of determining the species, recommended by the All-Russian Research Institute of Fisheries and Oceanography (VNIRO)*.
