A universal set of reagents for the isolation of DNA from biological material.
The COrDIS EXTRACT reagent kit is designed to obtain DNA preparations from human biological material for subsequent analysis by polymerase chain reaction.
Feature of the EXTRACT kit is a unique lysis buffer, providing highly efficient extraction of biomolecules from various carrier materials.
PRINCIPLE OF METHOD
In many cases, preliminary collection of the studied biological material is carried out using cotton or gauze swabs, probes and wands. The biological sample impregnates the fibers of the "media", which reduces its availability for subsequent exposure to lysing agents. This problem is especially acute when working with a low amount of the studied biomaterial (eg, when working with flushing contact biological traces or traces of biomaterial on clothing fragments). When DNA is isolated from tissue swabs, a significant part of the biomaterial remains inside the carrier fibers and is inaccessible for further research. This effect can be illustrated by measuring the concentration of DNA in the resulting preparations by real-time PCR.. No modern DNA isolation kit allows extraction of all biomaterial, collected on a cotton swab the first time. When DNA is re-isolated from the test swab, it is always possible to isolate additional residual DNA.
When creating "EXTRACT" a new unique set of detergents was used, allowing to increase the efficiency of extraction of biomaterial from fibers as natural, and synthetic fabric carriers.
The new EXTRACT lysis buffer allows achieving at least 4 times more efficient extraction of biomaterial, fiber-dried carrier materials in comparison with widely used foreign analogues.
The high efficiency of the primary extraction of biomaterial can significantly increase the effectiveness of the study of contact traces, washes of biomaterial from the scene, traces of biomaterial on fragments of clothing, sperm swabs, extractable by differential lysis protocol.
Further stages of isolation are carried out by DNA sorption on the surface of high-capacity sorbent magnetic granules. Isolated DNA can be used in molecular biological reactions without additional purification..
In addition to the standard allocation protocol, specialized protocols for extracting DNA from hair samples are provided, nails and subungual contents, paraffin blocks, mixed samples of epithelial and sperm components.
